Description: The purpose of this Funding Opportunity Announcement (FOA) is to encourage applications for projects to identify targets whose inhibition would induce synthetic lethality in cancers dependent on the expression of mutant KRas alleles. Despite recent advances in understanding the structural and biological complexities of mutant Ras proteins, the development of appropriate and effective methods to inhibit mutant Ras-dependent cancers has been hindered by the inability to directly target mutant Ras and the complexity of Ras processing, effector, and regulatory networks. Through this FOA, the NCI seeks to stimulate research that uses advanced or improved approaches for synthetic lethality screens that have greater specificity, reproducibility, and activity in physiologically relevant model systems than those used previously in first generation screening approaches. This FOA serves to engage extramural investigators in the mission of the NCI Frederick National Laboratory for Cancer Research (FNLCR) RAS Program to find ways to target Ras-dependent cancers. Investigators supported by this FOA will form a network to interact closely with the FNLCR RAS Program, actively exchange results across the network, and participate in cross-validation of potential targets identified by other awardees.
