This grant is for the renewal of the Multicenter Osteoarthritis Study (MOST) for another five-year funding period. MOST represents a uniquely valuable, large-scale observational cohort of 3026 individuals aged 50-79, either with knee osteoarthritis (OA) at baseline or at high risk of developing it. This pioneering study has prospectively examined knee OA development and progression concerning biomechanical factors and structural abnormalities. Over seven years, it has collected extensive repeated clinical, musculoskeletal, radiographic, and MRI data, alongside strength and physical performance measures, and plasma samples. The study also established a crucial specimen bank, facilitating future biochemical and genetic biomarker research in OA, thus significantly advancing understanding of this disease.

| Description: | The purpose of this limited competition FOA is to allow the Multicenter Osteoarthritis Study (MOST) investigators to submit a renewal application for another 5 year funding period, providing for resubmission and possible future revision applications. This is a uniquely valuable cohort of individuals who either had knee osteoarthritis (OA) at baseline or were at high risk of developing the disease because they had one or more disease risk factors. This is the first large scale observational study in knee OA to prospectively examine the development and progression of knee OA with respect to biomechanical factors and structural abnormalities. There is more than 7 years of repeated clinical and musculoskeletal evaluations, knee radiographs and knee MRI imaging, strength measures, physical performance measures and plasma samples in 3026 individuals who were 50-79 years at enrollment. The study also created a specimen bank for future biochemical and genetic studies of biomarkers in OA. |
